Route 159 is the northern extension of Main Street in Hartford, running along the west bank of the Connecticut River up to the Massachusetts state line in Agawam, Massachusetts. Massachusetts Route 159 starts at the state border and ends in Agawam's center.

Route 159 officially begins at the Hartford-Windsor town line as Windsor Avenue, near an interchange with Interstate 91. The road runs along the west bank of the Connecticut River, crossing Interstate 91 a second time and continuing into Massachusetts. It goes through the towns of Windsor, Windsor Locks, and Suffield.

In the 1920s, the route was known as State Highway 110. After the 1932 renumbering, it was designated as US 5A and remained that way until US 5A was decommissioned in 1970. The route was then redesignated as Route 159. [1]
