Pennsylvania Route 324 is a 12 mile long state highway located in the western part of Lancaster County, Pennsylvania. The southern terminus is at River Road (SR 3038) in Pequea and the northern terminus is at U.S. Route 222 in Lancaster. PA 324 is one of the few signed traffic routes in Pennsylvania that has at least one terminus at an unsigned road.

The route was signed in 1928. In the same year, the route underwent construction from Silver Mine Road to New Danville. This work was completed by the following year.
In 1936, the designation was moved off of the current Pennsylvania Route 741 alignment between PA 324 and US 222/Pennsylvania Route 272 and placed on its current alignment.
