Alkaline lysis

Alkaline lysis is a method used in molecular biology to isolate plasmid DNA from bacterial cells. Bacteria containing the plasmid of interest is first grown, then lysed with a strong alkaline solution made from sodium dodecyl sulfate (SDS) and sodium hydroxide. The alkaline punctures bacterial cell membranes. Through a series of suspension, precipitation, centrifuges, and the removal of supernatant, cellular debris is removed and the plasmid is isolated and purified. The protocol may vary slightly from lab to lab.
