An anonymous editor removed the note that DAPI stains nuclei in live cells; I restored the material.
DAPI and the Hoechst stains will both pass through the membranes of live cells—I've used them both in that exact application. The time and concentration of DAPI required vary depending on the cell line, but it will definitely cross an intact membrane. (The uptake is quite a bit faster for fixed, permeabilized cells, however.)
Googling around, it seems that a fairly high concentration is needed, and that it may not work for all cell lines; I add it to HeLa cells at about 10μg/mL in medium about 30 minutes prior to imaging. TenOfAllTrades(talk) 19:52, 6 March 2007 (UTC)
