Tilling (molecular biology)

Tilling (Targeting Induced Local Lesions in Genomes) is a method in molecular biology that allows a directed identification of mutations in a specific gene.
The method combines a standard technique, the mutagenesis with Ethyl methanesulfonate (EMS) with a new screening-technique that uses the possibility to identify Mismatch-Hybridisation with HPLC. By using the restriction enzyme Cel-I the method could be optimised.
Tilling was introduced in 2000, using the Arabidopsis-genome and is used for the mutagenesis of other organisms as well. Traditional mutagenesis-screens like the EMS mutagenesis can set point mutations, but the identification of these point mutations is much more difficult.
