Former Route 42 was one of the original Missouri state highways commissioned in 1922. Its eastern terminus was at Route 23 (now U.S. Route 67) south of Poplar Bluff; its western terminus is at Route 19 in Alton. It became part of U.S. Route 160 in the 1950s when that road was extended east to Poplar Bluff.
Missouri 42 was part of the original U.S. Route 62 when the U.S. Highway System was first planned, however when the number was changed to U.S. Route 60, the road took over Route 16.
