Mario is the hero of the Mushroom Kingdom who finds himself partnering up with his brother Luigi. Mario is the braver of the two. His abilities include Spin Jump, Mini Mario and the Firebrand. His battle moves include Swing Bros., Fire Bros., Chopper Bros. and Splash Bros.

Mario's younger brother, Luigi finds himself accidentally joining his brother on his quest. Luigi is more cowardly and easily frightens. Luigi's abilities are High Jump, Luigi Dunk and the Thunderhand.His battle move include Cyclone Bros., KnockBack Bros., Thunder Bros. and Bounce Bros.

Cackletta (ゲラゲモーナ Geragemona?) is an evil witch from the Beanbean Kingdom and the main villain the game. Aided by her sycophantic assistant Fawful, Cackletta attempted to steal Princess Peach's voice and use it to steal the Beanstar — a mystical object of the Beanbean Kingdom that can grant wishes. Beanbean lore states that only one with a beautiful voice can activate the Beanstar's mysterious powers to grant any wish — and Cackletta hopes to use this power to conquer Beanbean Kingdom and soon after, the Mushroom Kingdom. Cackletta's plans backfire, however, when she actually attempts to use Peach's voice. She finds that Toadsworth and the Mushroom retainers anticipated the ambush and switched Peach with Birdo, whom they dolled up like Peach. As a result, the Beanstar was infuriated by Birdo's squawking voice and did not grant Cackletta her wish.
Mario and Luigi soon caught up to Cackletta at Woohoo Hooniversity and fought her. She attacked ferociously, with black holes and lightning bolts, but, ultimately, was defeated by the Mario Bros. Her body was battered up so badly, she could no longer move around on her own, and was close to her turn in death. Fawful absorbed Cackletta's spirit into a vacuum cleaner-like device to save her and tried to attack the Mario bros. He was stopped by Prince Peasley, who shot them out of the Hooniversity. Bowser was suffering from amnesia for some time after being shot out of a large cannon and was wandering idly around Beanbean Kingdom. Just when he remembered who he was, Fawful met him and sent Cackletta's spirit into his body. Cackletta promptly renamed herself "Bowletta", a combination of Bowser's name and hers.
Armed with the new alias, Bowletta continued her plan to obtain the Beanstar and Peach's voice. The Mario Bros. eventually caught up with Bowletta, and after fighting through the notoriously spoiled Koopalings (Bowser's malicious children), and Fawful, they fought Bowletta. Bowletta gave up without really giving a fight, and it seemed too easy, until Bowletta swallowed them whole. Inside Bowser's stomach, Mario and Luigi encountered Cackletta's spirit inside of the body. After defeating the specter, it left Bowser's body, returning him to normal.

Fawful (ゲラコビッツ Gerakobittsu?) is a fictional video game character who appears in the game Mario & Luigi: Superstar Saga and its sequel, Mario & Luigi: Partners in Time. He is the accomplice to the villainous Cackletta and a source of the game's comic relief.
Fawful has found fame as one of the more quotable characters in the game, as he has a bizarre grasp of the English language, turning basic sentences into nonsense. In fact some consider his speech patterns to be one of the game's many in-jokes (in this case, a reference to the days of the NES, when translation was poor; see Engrish). For example, he shouts "Fink-rat! Have you readiness for this?" in his first battle with the Mario Brothers. His catchphrase is "I HAVE FURY!"
Fawful returns in a cameo appearance in Mario & Luigi: Partners in Time, thus proving that Fawful did, indeed, survive what happened back in Superstar Saga. He is now a merchant below Peach's castle in the present. The shop can be accessed by tunnelling under the pipes as the babies. Here, he will trade beans for valuable badges.

Popple is a recurring enemy who Mario and Luigi encounter and battle through various points in the game. He is a thief in constant search of a dependable sidekick. He initially recruits Bowser (referred to as "Rookie" by Popple), and later fights Mario and Luigi alone before recruiting Birdo as his partner in his final battle against the Mario Bros. He has a tendency to blame his partners for their joint defeats, which the amnesiac Bowser takes but Birdo refuses to. After being knocked flying by Birdo he appears in Little Fungitown being forced to do a young toad's cleaning chores and being called Rookie (he might have lost his own memory like his first rookie, Bowser, had).
