New York State Route 21 is a state highway in the western part of New York State. The northern terminus is at New York State Route 104 in Wayne County. The southern terminus is at New York State Route 417 in Allegany County.

In 1980, NY 21's northern terminus was moved southward from the intersection with Lake Road (formerly New York State Route 18) in Pultneyville to its present location at the intersection with NY 104 in Williamson.
Before the 1930 renumbering, the number 21 was assigned to the route from Poughkeepsie to Amenia (present day U.S. Route 44).
