New York State Route 376 is a New York state highway located entirely within Dutchess County. The route travels from Poughkeepsie, where it meets U.S. Route 44 and New York State Route 55, passing south through Red Oaks Mill and Hopewell Junction, ending in East Fishkill where it meets New York State Route 52.

Its exact route has changed a few times over the years, but NY-376 currently begins at NY-52 in Hopewell Junction and heads north to NY-82 where a short 30-foot duplex is formed. The road then heads north, then leaves 82 to go east (signed as north), which are two of six intersections on 376 requiring a turn for through traffic. After that duplex, one will encounter a sharp 90ยบ S-Curve, on which the speed limit is 5 MPH. Soon after, one will find another curve, but at 20 MPH. After this, the speed limit returns to 45 before meeting CR-29 at which point 376 heads west, then back north at CR-104. The route also meets NY-113 and CR-77 at the same intersection. Later, 376 meets US-44/NY-55 at it's northern terminus.
