Season 9 ended with Mo Collins and Josh Meyers leaving the cast to pursue movie careers. That meant last year's featured cast members - Daniele Gaither, Keegan-Michael Key, Nicole Parker, and Jordan Peele - would be upgraded to repertory players. And the timing couldn't have been better. After the Ashlee Simpson incident on Saturday Night Live, Nicole Parker would frequently appear in sketches as the pop singer. Outside of that, she would also impress audiences with her other impersonations of Ellen DeGeneres, Kathy Griffin, and Britney Spears.
African-American newcomers Gaither, Key, and Peele would further cement their place on MadTV. The latter two would do a repertory sketch as the "Man Up" crew who'd have to break jinxes. Gaither would be featured in plenty of sketches, and would be praised as a great scene stealer.
The season was also the last for Ron Pederson and Paul Vogt. While they were funny, they had budget cuts and were no longer needed. Regardless, Pederson entertained audiences with his Clay Aiken impersonation, and Vogt owned a lot of funny sketches, such as the Fanta commercial parody as Beth, Lillian Verner Game Show as Dale Priskett, and Inside the Actor's Studio as James Lipton. Additionally, Aries Spears, who was a MADtv cast member since Season 3, would leave the show at the end of the season, tying Debra Wilson's then record for the longest tenure for a MADtv cast member (it would later be surpassed by Michael McDonald's nine years).
On the subject of new cast members, Crista Flanagan would join the cast, but not be updated to repertory player status until next season. Regardless, she brought a preview of what to expect around that time, including very demure stand-up comic Luann Lockheart.
The 10th season would also see former cast members make returns. Will Sasso returned in 2004 to reprise his role as Kenny Rogers in a reality show-themed sketch. Mo Collins returned for episode #1014 (along with Susan Sarandon, her first appearance since February 10, 2001) to reprise famous roles such as Doreen Larkin and Lorraine Swanson. Artie Lange also returned for the second time in a row in two episodes. Also Alex Borstein reprised her role as Miss Swan, and joined her Family Guy co-star Seth Green once again for another Brightling sketch. Finally, original players Nicole Sullivan and Debra Wilson (for the first time since she left in 2003) made big waves when they returned, reprising some of their famous characters.
Staff writer Steven Cragg appeared in three episodes starring in self-made short films.

The title sequence starts off with the MADtv logo appears across the skyline of Los Angeles which takes up of the whole screen. The theme song, which is performed by the hip-hop group Heavy D & the Boyz, begins and an announcer announces "It's MADtv! Starring...," and then he introduces (alphabetically) the name of each repertory cast member, followed by the featured cast. As he does so, the screen dissolves into three live action clips (all of the same clip) of an individual cast member. Then the three screens multiple until it takes up the whole screen. Then all of the multiple clips flip over and displays another clip of the same cast member. As the multiple clips are reduced to one clip, a still color photo of the cast member is super imposed on the screen with his/her name appearing in caption over the photo. When the last cast member/guest is introduced, the music stops and the title sequence ends with the phrase "You are now watching MADtv..."

Joan of Arcadia actress Amber Tamblyn plays a teen who helps celebrate average Asian Hideki's (Bobby Lee) birthday. Gray (Jordan Peele) and Carl (Keegan-Michael Key) try to break hexes before performing MacBeth on stage. Sean Gidcomb (Michael McDonald) prohibits employees from relationships within the workplace. A 1950s film promotes tolerance towards African-American neighbors, but the message goes haywire when one of them wants to date a white family's daughter (Nicole Parker). - This was the last season to feature Aries Spears, who was a MADtv cast member since Season 3. Spears' eight year tenure tied Debra Wilson record for the longest tenure for a MADtv cast member. Only Michael McDonald would surpass such a long tenure, at the start of the twelfth season.
- This was also the last season for Ron Pederson and Paul Vogt. They were fired because of budget cuts, and the producers did not see any need to keep them on the show any longer.
- Out of the two featured players of Season 10, only Crista Flannagan was retained and promoted to repertory status for the following season. Spencer Kayden's contract was not renewed.
- This season marked MADtv's tenth anniversary, even it wasnt officialy 10 years until mid-October of 2005. As such, many former cast members, like Artie Lange, Nicole Sullivan, Debra Wilson, and Will Sasso, returned to film special sketches which were broadcast throughout the season.

As of March 2007, there have been no video or DVD release of all of the MADtv episodes from Season Ten. However, several sketches from Season Ten was featured on the The Best of MADtv Seasons 8, 9 & 10 which was released on October 25, 2005. The Best of MADtv Seasons 8, 9 & 10 DVD sketch material includes celebrity impersonations, music video, commercial, movie, and TV show parodies, recurring characters, musical guests, and the occasional mock cartoons.
